An Open-label, Multicenter, Randomized Phase 2 Study of the ATR Inhibitor Tuvusertib in Combination With the PARP Inhibitor Niraparib or the ATM Inhibitor Lartesertib in Participants With BRCA Mutant and/or Homologous Recombination deficiency (HRD)-Positive Epithelial Ovarian Cancer That Progressed on Prior PARP Inhibitor Therapy

You maybe eligible to take part in this trial if you:
- Are aged 18 years and over
- Have high-grade serous or high-grade endometrioid ovarian, primary peritoneal, and/or fallopian tube cancer that is recurrent
- Have a BRCA mutation or positive HRD status
- Have disease progression confirmed by scan whilst taking a parp inhibitor
- Have taken the parp inhibitor for at least six months before the disease progressed, and taken it in either the first or second line setting
- Have disease on scan that can be measured
- Are in good general physical health

About the trial
This trial is for those with a diagnosis of high grade serous or endometrioid ovarian cancer that has progressed while on a Parp inhibitor (Olaparib, Niraparib or Rucaparib). It is an open label study, which means both you and the doctor will know what treatment you are receiving.
The researchers conducting the trial are testing a drug called Tuversertib in combination with other drugs, Niraparib and Lartesertib.
Tuversertib and Lartesertib are drugs designed to block ATR and ATM proteins from working; these proteins are involved with DNA repair. When the DNA in cancer cells is damaged and their repair mechanism is blocked by the trial drugs then the cancer cells will die.The study will look at whether the cancer disappears, shrinks, remains the same or gets worse on the treatment. The study will also look at the safety and side effects of the treatment to help plan the second part of the trial.
Niraparib is a parp inhibitor.
The drugs being tested in Part A of this trial are:
Tuvusertib – an ATR inhibitor (tablet taken by mouth)
Lartesertib – an ATM inhibitor (tablet taken by mouth)
DDRiver has 2 different treatment arms and if you take part, you will be randomised (placed into a treatment group by a computer) into one of the treatment groups.
Arm 1 – Tuvusertib +Niraparib (parp inhibitor; tablet taken by mouth)
Arm 2 – Tuvusertib + Lartesertib
All participants will be followed up by the research team.
Part B of this trial is due to commence later in the year and will be looking at comparing specific doses of the drugs used in this trial alongside Tuvusertib given on its own.
